Tumor Necrosis Factor as a Marker for Determining the Pharmacological Activity of NSAIDs and Paracetamol in Case of Hypothyroidism and Osteoarthritis

Abstract

Recently the problems of comorbid pathology have received increasing attention, in particular the issues of development and effective treatment of comorbidity with hypothyroidism were studied, however, the effects and interactions of NSAIDs and paracetamol in the treatment of osteoarthritis on the background of hypothyroidism were not thoroughly studied. The purpose of the study was to research the pharmacological activity of NSAIDs and paracetamol on the level of tumor necrosis factor in the serum of rats with experimental equivalents of hypothyroidism and osteoarthritis. Material and methods. The study was conducted on 80 white non-linear rats of both sexes, weighing 200-250 g, which were kept under standard conditions of the vivarium of the State Institution “DMA of the Ministry of Health of Ukraine”. Experimental osteoarthritis was reproduced through a single intraarticular administration of 0.1 ml of a solution monoiodacetic acid into the knee joint, which was prepared at the rate of 3 mg of reagent to 50 µl of sterile saline. Experimental hypothyroidism was reproduced by enteral introduction of a 0.02% solution carbimazole (the drug "Espa-carb", manufactured by Esparma GmbH, Germany; in tablets of 5 or 10 mg), which was prepared at the rate of 5 mg in 250 ml saline and given with drinking diet for 6 weeks. The drugs were administered daily from day 42 of the experiment on the background of increasing pathological changes within 5 days. To obtain a homogeneous suspension for intragastric administration tablets used solution of Tween-80 (Polysorbate 80, Ukraine). Quantitative levels of tumor necrosis factor in the serum was determined by competitive ELISA in vitro twice (42 and 47 days of the experiment) using the enzyme immunoassay system MBS (USA) according to the method of the manufacturer. Statistical data processing was performed using the STATISTICA 6.1 software package (StatSoftInc., Serial number AGAR909E415822FA). Conclusion. The author found that the levels of tumor necrosis factor allowed evaluating the activity of inflammatory process on the background of experimental equivalents of osteoarthritis and hypothyroidism. The obtained data of the contents of tumor necrosis factor in the serum of rats reflected the degree of influence of NSAIDs and paracetamol on the activity of the inflammatory process due to the interaction of drugs in experimental osteoarthritis and hypothyroidism. The degree of impact on inflammatory processes in the joints with the help of studied drugs can be arranged as follows: diclofenac > ibuprofen > nimesulide = meloxicam > celecoxib > paracetamol.
